Objective: In this study, we observed the effects of Tiao Chang Xiao Yan Tablet on colon mucosal damage index, pro-injiammatory factors IL-1 beta and IL-6, the anti-inflammatory factors IL-13,1L-4 and utophagy protein LC3B (LC3-II) in ulcerative Colitis rats. Methods: The 42 male Wistar rats were randomly divided into 6 groups: normal control group, model group, Tiao Chang Xiao Yan Tablet high dove group, medium dose group, low dose group, mesalazine group. TNBS-40% ethanol method was used to prepare model of chronic recurrent UC. The high, medium and low dose groups were given Tiao Chang Xiao Yan Tablet according to the corresponding dose for intragastric administration; SASP group was given SASP aqueous solution for intragastric administration; mesalazine group was given mesalazine queous solution for intragastric administration; model group and Normal control group was given saline lavage for intragastric administration. Each group was treated for 8 days, then observed the gross morphology of colon tissue in rats. Me mucosa damage index (CMDI) was eveluaated. The levels of 1L-1 beta, 1L-4, IL-6 and IL-13 of serum inflammatory factors were detected by ELISA enzyme immunoassay. The level of LC3B (LC3-II) in colon tissue was detected by immunohistochemical method. Results: It can see colon mucosal ulcer, and wall congestion and edema intestinal in model group. Colon mucosal damage index (CMDI) in model group was significantly higher than that in normal group (P<0.05). Pathology also showed that colonic mucosa or muscular layer were defected, with a large number of inflammatory cell infiltration, even severe transmural inflammation can be foul. All above that confirmed the success of the UC model. After intervention with Tiaochartg Xiaoyan Tablet and Mesalazine, CMDI was lower than that in model group (P<0.0.5), and the degree of CMDI reduction was dose-dependent with Tiao Chang Xiao Yan Tablet. Pathology also showed that inflammatory cells were reduced, and ulcers were repaired in varying degrees. The pro-inflammatory factors 1L-1 beta and IL-6 of model group were higher (P<0.05), and the anti-inflammatory factors IL-13 were lower (P<0.05) than those in normal control group After the intervention of Tiaochang Xiaoyan Tablet and Mesalazine, the pro-inflammatory factors IL-1 beta and 1L-6 decreased (P<0.05), and the anti-inflammatory factors IL-13 increased ( P<0.05). The expression of LC3B in colon tissue was higher than normal group, After the intervention of Tiaorhang Xiaoyan Tablet and Mesalazine, LC3B (LC3-II) decreased (P<0.05). Conclusion: Tiao Chang Xiao Yan Tablet is effective in the treatment of chronic reccurent ulcerative colitis rats. Its mechanism may be that Tiao Chang Xiao Yan Tablet can balance the pro-inflammatory factors and anti-inflammatory factors, inhibit autophagy, then promote the repair of colonic ulcer.
